The LSI IGH/MYC, CEP 8 Tri-color, Dual Fusion Translocation Probe is designed to detect the juxtaposition of immunoglobulin heavy chain (IGH) locus and MYC gene region sequences. Inclusion of the SpectrumAqua CEP 8 in this probe mixture serves as an indicator for chromosome 8 which is important when MYC amplification and loss of the der(8) chromosome occurs. The translocation t(8;14) (q24;q32) involving IGH at 14q32 and the MYC region at 8q24 is the most frequently observed MYC region translocation. The remaining cases appear to have one of the variant MYC translocations, either t(8;22)(q24;q11) or t(2;8)(p11;q24) that involve one of the two light chain immunoglobulin loci (kappa on chromosome 2 or lambda on 22). These three translocations are believed to disrupt the normal regulation of the C-MYC transcription factor by bringing the MYC gene under control of a regulatory element from one of the immunoglobulin loci.

The LSI IGH/MYC, CEP 8 Tri-color, Dual Fusion Translocation Probe is a mixture of a 1.5 Mb SpectrumGreen labeled LSI IGH probe, an 821 kb SpectrumOrange labeled LSI MYC probe and the SpectrumAqua labeled CEP 8 probe. The IGH probe contains sequences homologous to essentially the entire IGH locus as well as sequences extending about 300 kb beyond the 3' end of the IGH locus. The large MYC probe extends 385 kb upstream of MYC and 431 kb 3' beyond MYC. The CEP 8 probe targets the alpha satellite sequences on chromosome 8.
