The LSI PML/RARA Dual Color Translocation Probe targets t(15;17). The t(15;17) involving the PML (15q22) and RARA (17q21) genes results in the formation of the PML/RARA gene fusion product. The t(15;17) breakpoint on chromosome 15 occurs in a 13 kb region of the PML gene. There are three breakpoint cluster regions:
  • bcr 3 : spans from intron 3 through the 5' end of intron 4
  • bcr 2: extends from exon 5 to exon 6
  • bcr 1: includes intron 6 and extends into exon 7
The majority of breaks occur in bcr 1, followed by bcr 3 and bcr 2. The break-point on chromosome 17 occurs within the approximately 17 kb intron 2 of the RARA gene.

The LSI PML/RARA Dual Color Translocation Probe is a mixture of the LSI PML probe labeled with SpectrumOrange and the LSI RARA probe labeled with SpectrumGreen. The PML probe covers the region that begins at intron 7, a span of 5.8 kb, and extends toward the centromere of chromosome 15 for approximately 180 kb. The RARA probe begins about 500 bp 3' to exon 6 and extends approximately 400 kb toward the telomere of chromosome 17.

Results of Hybridization


In a normal cell, the expected pattern for a nucleus hybridized with the LSI PML/RARA probe is a two orange and two green (2O2G) signal pattern.
