Metaphase slides prepared from normal PHA-stimulated peripheral blood lymphocytes for optimum in situ hybridization results in CGH.

This control DNA is extracted from the MPE-600 tumor cell line that has known chromosome aberrations determined from CGH and karyotype analysis. Expected CGH results for the control DNA are described to provide control data for evaluation of hybridization efficiency of CGH techniques.
This product is provided as positive control DNA for CGH. The DNA is extracted from the MPE-600 tumor cell line that has known chromosome aberrations determined from CGH and karyotype analysis. The DNA is directly labeled with SpectrumGreen fluorophore.
Controls may be useful as comparison for fluorophore labeling and hybridization results.

CGH Nick Translation Kit With Control DNA (MPE 600)
The CGH Nick Translation Kit is used to label DNA (that is isolated for CGH) with either SpectrumGreen or SpectrumRed™ fluorophore.
Components in the CGH Nick Translation Kit include:
- Two positive controls
- Unlabeled DNA (3 µg) extracted from the MPE-600 cell line
- SpectrumGreen labeled MPE-600 DNA (1.5 µg)
- Nick translation enzyme mix (525 µL)
- 10X Nick translation buffer (300 µL)
- dATP, dCTP, dGTP, and dTTP solutions (200 µL each)
- Nuclease-free water (1500 µL)
- Easy-to-follow protocol

CGH Hybridization Reagents are used to prepare a CGH hybridization following probe preparation.
Hybridization Reagents include:
- CGH Hybridization Buffer, 250 ìL
- 20X SSC, 66 g
- NP-40, 1000 ìL
- DAPI II Counterstain with antifade, 250 ìL
- CGH protocol
COT-1 blocking DNA for use in FISH/CGH.

Labeled Reference DNA:
Ready-to-use fluorophore-labeled female total human genomic DNA is isolated from normal lymphocytes for use in CGH. The reagent does not contain COT-1® DNA. The DNA was extracted from the peripheral blood of karyotypically normal female individuals and contains minimal polymorphic regions.
